Instructions

A copy of Ristocetin Cofactor, von Willebrand Factor Antigen, and Factor VIII Activity are required to be sent with the specimen. Multimer analysis will only be performed when Ristocetin Cofactor or von Willebrand Factor Antigen are below normal.

Specimen Collection Criteria

Collect: One Light Blue-top Sodium Citrate tubes. Mix gently by inverting 3-4 times.

TUBES MUST BE FULL.

Physician Office/Draw Specimen Preparation

Centrifuge 15 minutes at 1500 g within one hour of collection. Using a plastic pipette, remove plasma, taking care to avoid the WBC/platelet buffy layer and place into a plastic vial. Centrifuge a second time and transfer platelet-poor plasma into a new plastic vial(s). Freeze (-20°C/-4°F or below)  immediately and transport.

Preparation for Courier Transport

Transport: 1.0 mL platelet poor plasma (PPP) aliquot, frozen (-20°C/-4°F or below). (Minimum: 0.2 mL)

Rejection Criteria

  • Grossly hemolyzed specimens.
  • Grossly icteric specimens.
  • Grossly lipemic specimens.
  • Specimens not collected and processed as indicated.

In-Lab Processing

Specimen Stability for Testing:

Room Temperature (20-26°C or 68-78.8°F): Unacceptable
Refrigerated (2-8°C or 36-46°F): Unacceptable
Frozen (-20°C/-4°F or below): 30 days

Specimen Storage in Department Prior to Disposal:

Specimen retention time is determined by the policy of the reference laboratory. Contact the Send Outs Laboratory with any questions.

    Clinical Utility

    von Willebrand Antigen, Multimeric – von Willebrand Disease is the most common hereditary bleeding disorder and it may also be acquired. Von Willebrand Factor (VWF) is critical for normal hemostasis; mediating platelet adhesion to the damaged endothelium and protecting Factor VIII from proteolytic degradation in circulation. VWF Multimeric Analysis is used to assist in categorizing disease subtype.
