Specimen Collection Criteria

Collect: Amniotic fluid, Cerebrospinal fluid (CSF), Ocular fluid, or biopsy tissue in a sterile container.

Specimen source is required.

Physician Office/Draw Specimen Preparation

Fluid: Maintain specimens in a sterile container and refrigerate (2-8°C or 36-46°F).

Tissue: Place specimen in M4 Transport and refrigerate (2-8°C or 36-46°F).

Preparation for Courier Transport

Transport: 0.5 mL amniotic fluid or CSF refrigerated (2-8°C or 36-46°F), or 0.3 mL ocular fluid refrigerated (2-8°C or 36-46°F), or tissue in M4 transport tube refrigerated (2-8°C or 36-46°F).

Rejection Criteria

Specimens not collected and processed as indicated.

In-Lab Processing

Transport: 0.5 mL amniotic fluid or CSF refrigerated (2-8°C or 36-46°F), or 0.3 mL ocular fluid refrigerated (2-8°C or 36-46°F), or tissue in M4 transport tube refrigerated (2-8°C or 36-46°F).

Storage

Specimen Stability for Testing:

Room temperature (20-26°C or 68-78.8°F): Unacceptable
Refrigerated (2-8°C or 36-46°F): 7 days
Frozen (-20°C/-4°F or below): 7 days

Specimen retention time is determined by the policy of the reference laboratory. Contact the Send Outs Laboratory with any questions.

Test Methodology

Qualitative Polymerase Chain Reaction (PCR).

Interpretation

A negative result does not rule out the presence of PCR inhibitors in the patient specimen. Toxoplasma gondii DNA concentration below the level of detection by the assay.

Epidemiology

Toxoplasmosis occurs worldwide. Asymptomatic infections are common. Serological surveys show that 7 to 80% of various populations are infected. The developing fetus and immunocompromised patients are at risk for severe disease (1).
