Instructions

This assay includes:

Specimen Collection Criteria

Collect: One Gold-top SST tube.
Also acceptable: One plain Red-top tube.

Physician Office/Draw Specimen Preparation

Let specimen clot 30-60 minutes then centrifuge to separate serum from cells. Transfer serum to a plastic transport tube and maintain at room temperature (20-26°C or 68-78.8°F) prior to transport.

Preparation for Courier Transport

Transport: 2.0 mL serum, maintain at room temperature (20-26°C or 68-78.8°F) prior to transport. (Minimum: 1.0 mL)

Rejection Criteria

  • Other body fluids
  • Contaminated specimens
  • Hemolyzed specimens
  • Icteric specimens
  • Lipemic specimens
  • Specimens not collected and processed as indicated.

In-Lab Processing

Let specimen clot 30-60 minutes then centrifuge to separate serum from cells. Transfer serum to a plastic transport tube and maintain at room temperature (20-26°C or 68-78.8°F) prior to transport.

Test Methodology

Immunodiffusion (ID).

Clinical Utility

Fungal Panel 1, Immunodiffusion – This immunodiffusion assay is highly specific for histoplasmosis infection but may have low sensitivity in early infection. Two different immunoprecipitin bands may be detected. The M band develops with acute infection, is often present in chronic infection, and may persist for months to years. The H band, which is usually present with the M band, generally suggests a chronic or severe acute infection. The presence of both the H and M bands is considered conclusive for the diagnosis of histoplasmosis.
